TSOD mouse

TSOD mice spontaneously develop polygenic obesity with non-alcoholic fatty liver disease that progresses to steatohepatitis. Livers display mixed micro- and macrovesicular steatosis, hepatocyte ballooning, Mallory-Denk bodies, inflammatory infiltration, fibrosis, and eventual tumor development; elevated hepatic PPAR-gamma suggests excessive lipogenesis as a contributing factor.

Outcome measures

Biological characteristics

Phenotype stage
Steatosis and steatohepatitis features are established before 1 year of age; inflammatory infiltration, fibrosis and tumor development are reported by 1 year of age.
Research questions
Used as a polygenic model of NAFLD with obesity that recapitulates the natural course from steatosis through steatohepatitis, fibrosis and tumor development.
Limitations
The evidence adopted here for TSOD comes from a review citing the original TSOD characterization literature rather than from direct use of the model in the primary studies of this package; TSOD mice develop polygenic obesity and the specific alleles underlying the NAFLD phenotype are not defined; spontaneous tumor development after 1 year of age limits use of the model for interventional studies that require a non-tumor endpoint; genetic background substrain and precise allele composition are not reported in the sources adopted here.

Experimental methods

Experimental protocols

  • Hematoxylin and Eosin (H&E) staining for histopathology

Hematoxylin and eosin (H&E) staining of fixed, paraffin-embedded tissue sections (4 μm). The workflow covers dewaxing, rehydration, hematoxylin nuclear staining, bluing, eosin counterstaining, dehydration and clearing, and coverslipping, yielding blue-purple nuclei and pink cytoplasm and connective tissue fibers for light-microscopic assessment of cellular and tissue morphology.
