Procedure
Prepare the RNA and primer mixture
In a sterile centrifuge tube, combine RNA (1ng-1 µg total RNA or 1-100 ng poly(A) RNA), 2 µl primer (50 µM dT or 60 µM Random Primer Mix), and 1 µl 10 mM dNTP mix. Bring to 12 µl with nuclease-free water.
RNA denaturation
The mixture may be heated at 65 °C - 70 °C for 3-5 minutes, briefly centrifuged, and immediately placed on ice.
Prepare the reverse transcription reaction
Add 4 µl 5X ProtoScript II RT Reaction buffer, 2 µl 0.1 M DTT, 1 µl Murine RNase inhibitor (40 U/µl), and 1 µl ProtoScript II Reverse Transcriptase (200 U/µl), giving a total volume of 20 µl.
Random-primer annealing
When using Random Primer Mix, incubate at 25 °C for 00:05:00 before the reaction at 42 °C.
Reverse transcription
Incubate at 42 °C for 30 minutes-1 hour.
Enzyme inactivation
Incubate at 80 °C for 00:05:00.
Store cDNA
Store the product at -20 °C or proceed to the subsequent assay.
Quality control and readouts
Use intact RNA free of RNase contamination; record the input amount for each sample and check no-reverse-transcription controls. Use consistent cDNA input amounts for subsequent quantification.
First-strand cDNA for subsequent amplification of target sequences.
